Can the Supreme Court Justices be tested on their overall comprehension of the US Constitution?

No---but the Congress has power to revise the Judiciary---also the SCOTUS has no authority to intrepret the Constitution but can decide cases they choose to hear---therein lies the failings of the SCOTUS---if they do not want to rule they simply refuse to hear a case.
